Kind of unsure of how this would work.

If you use Giant to get a Ghost in grave, can you use Ghost's effect to get Golem too?

 

Giant would special Ghost in defense, and since Ghost would turn to defense after its effect would the effect still happen?

Yes, that is legal. Ghost's effect cannot miss the timing.

I think the question was more along the lines of if it can summon a monster but not change to defense.

According to the Conjunction Functions article, "then" means resolve as much as possible, but the second is not required for the first to occur.
